If Suppliers A and B instances are synced with our instance, can a Supplier A raise a ticket and assign it to Supplier B easily or do we need to manually assign the ticket when it hits our instance?

You can define a trigger which will sent the issue to SupplierB.
For instance if the issue contains a component ‘MadeBySupplierB’, the trigger can then send it over there.
